Separation of DNA fragments by cross-linked polymer coating capillary electrophoresis with non-gel sieving matrix

A new type of flush/fill equipment driven by high-pressure (1MPa) nitrogen gas for filling the capillary is demonstrated. High-viscous and microlitre grade volume solution can be filled into a capillary by means of this equipment. The capillary coated by cross-linked polyacrylamide and filled with “interpenetrating polymer networks” non-gels sieving matrix (0.8% hydroxyethyl cellulose, 10% sucrose in 1×TBE buffer) was applied for separating DNA fragments. All 11 fragments of Ø174/Hae III DNA were successfully separated. The man-made circle plasmid from E. coli were separated and detected.     

重组杆状病毒的研究——Ⅱ.带标记的多角体转移载体质粒的构建

用大肠杆菌β-半乳糖苷酶基因,杆状病毒ETL启动子和含多角体基因的病毒DNA片段构建了多角体转移载体质粒pEZGL,还对其特点进行了讨论。     

利福平对大肠杆菌F′质粒的消除

研究了利福平(rif)对大肠杆菌F′质粒的消除效应.实验结果表明,消除质粒以rif的浓度50—100μg/mL为宜.通过选择培养,在LB和LB+cm平板上,就能选出F~-菌株,因F~+抗cm,E~-不抗cm.选得的F~-菌株是:E.Coli XAC F~-/△(lac pro)nal.     

A human homolog of the yeast gene encoding tRNA 2′-phosphotransferase: cloning,characterization and complementation analysis

The Saccharomyces cerevisiae TPT1 gene plays a role in removing the 2-phosphate from ligated tRNA during the maturation of pre-tRNA. Here we reported the cloning and characterization of the human TRPT1 gene as a homolog of yeast TPT1. The TRPT1 gene is located at human chromosome 11q13 and encodes a polypeptide of 253 amino acids. BLAST searches with its amino acid sequence revealed the ubiquitous occurrence of TRPT1 homologs and their functional relationships with the presence of the DUF60/KptA domain. Northern analysis demonstrated that the gene is primarily expressed in heart and skeletal muscle, with lower or undetectable levels in other tissues studied. 含副溶血弧菌(Vibrio parahaemolyticus)FlaI基因真核表达重组质粒的构建

用ClaI／EcoRI双切酶切带有副溶血弧菌（Vibrio parahaemolyticus)极鞭毛蛋白FlaI基因的质粒p MD18-FlaI，回收目的基因片段，插和到真核细胞表达载体pIRESneo同样经ClaI/EcoRI切开的窗口中，成功地将FlaI克隆到pIRESneo中，获得有意义的重组质粒pIREneo－FlaI,构建的pIREneo－FlaI真核表达重组质粒，将为海水经济鱼类的副溶血弧菌DNA疫苗的研制奠定基础。     

两种氰污染环境降氰菌的类群研究

本文对昆明大板桥煤气厂污水处理站各处理环节的污水及镇沅金矿３个污水样点中的细菌数量，类群进行研究、比较，筛选高效降氰菌株，并对这两种不同的氰污染环境中的细菌进行质粒检测．     

基因工程菌的发酵条件研究

为了获得商品化基因工程产品，要求所选用的基因工程菌高效表达重组蛋白．然而，随着发酵规模的扩大，诸多因素影响基因工程菌蛋白的表达．成功的大规模发酵有两个问题是重要的．一是质粒的稳定性，二是高密度发酵．本文从理论上阐明了ｐＨ，温度、溶氧、比生长速率等条件对重组蛋白产生影响的研究进展．     

小肠结肠炎耶氏菌体外毒力试验

为了初步研究小肠结肠炎耶氏菌（Ｙ．ｅ．）致病机理，本文报告应用依钙、自凝、刚果红平板毒力试验和质粒ＤＮＡ提取方法，进行Ｙ．ｅ．体外毒力探讨。结果表明：带毒力质粒的Ｙ．ｅ．菌株Ｇ１５１，９２０和ＬＡＢ－Ｂ１８２依钙、自凝和刚果红平板毒力试验阳性；不带毒力质粒的Ｙ．ｅ．菌株８２－１４０和４３９－８０Ｖ￣－依钙、自凝和刚果红平板毒力试验阴性。依钙、自凝、刚果红平板毒力试验阳性与携带毒力质粒的一致性以及依钙、自凝、刚果红平板毒力试验阴性与不携带毒力质粒的一致性表明：Ｙ．ｅ．的依钙、自凝和吸收刚果红特性是毒力质粒编码。
